PSVI-2 Relationship of Sperm Plasma Serine Protease Inhibitor (Serpina5) and Dystroglycan (DAG1) Proteins with Dairy Bull Fertility

چکیده
Abstract It has been reported that plasma serine protease inhibitor (SERPINA5) and dystroglycan (DAG1) are loosely attached to the sperm SERPINA5 can also be present on tails. The objective of this study was evaluate whether and/or DAG1 could used as a fertility marker in dairy bulls. Frozen semen from bulls (n=19) were evaluated for concentration percentage tail labeled (SERPINA5-tail) by immunofluorescence minimum 200 cells per bull two different ejaculates. Semen total motility, progressive viability. vitro embryo production (~150 oocytes/bull). Bull classified methods, sire conception rates [SCR - High-SCR ( >1.0) or Low-SCR (< -4.0)] ratio embryos developed into blastocyst (BL) those had cleaved (CL) Good (BL/CL≥38.5%) Poor (BL/CL< 38.5%). GLIMMIX procedure SAS classification with SCR, BL/CL, interaction model random effect. Progressive CL, concentration, SERPINA5-tail did not differ (P≥0.14) among SCR BL/CL classifications their interaction. There no difference (P>0.33) motility between classification; however, significant (P=0.02; High-SCR/Good 54.9±2.8%, High-SCR/Poor 39.4±5.4%, Low-SCR/Good 44.8±4.5%, Low-SCR/Poor 52.03±3.2%). decreased BL (P=0.03; 30.6±1.6%, 35.8±1.4%, respectively) tended have (P=0.10; 38.1±2.0%, 42.9±1.8%, compared In summary, do seem putative marker. is good predictor since may developments.

Bull Fertility and Its Relation with Density Gradient Selected Sperm
Background sperm selection is generally used to describe methods for separation of spermatozoa for in-vitro-assisted reproduction. Few studies reported relation in vivo fertility and semen parameters after selection of fertile spermatozoa; hence, our research attempt to assess different semen parameters after post-thaw (PT) or after selecting sperm sample (SSp), using density gradient separatio...
متن کاملRelationship of bull fertility to sperm nuclear shape.
The relationship between sperm nuclear shape and bull fertility was determined. Two groups of bulls, 3 per group, were selected. Bulls differed in fertility based on lifetime nonreturn rates. Digital images of propidium iodide-stained sperm from each bull were collected and shape-evaluated by Fourier harmonic amplitudes 0 to 5. A discriminant function (P < .05) was constructed based on harmonic...
متن کاملNematode sperm maturation triggered by protease involves sperm-secreted serine protease inhibitor (Serpin).
Spermiogenesis is a series of poorly understood morphological, physiological and biochemical processes that occur during the transition of immotile spermatids into motile, fertilization-competent spermatozoa. Here, we identified a Serpin (serine protease inhibitor) family protein (As_SRP-1) that is secreted from spermatids during nematode Ascaris suum spermiogenesis (also called sperm activatio...
متن کاملSperm Characteristics in Cryopreserved Buffalo Bull Semen and Field Fertility
Semen quality and its relationship to fertility are major concern in animal production. The aim of this study was to assess the relationship between frozen thawed sperm characteristics and fertility following in vivo fertility through Artificial Insemination(AI). Semen samples were collected from four buffalo bulls. Semen volume and sperm concentration appeared to be significantly different (P0...
